dc.description.abstract Background: As the number of cases of coronavirus disease (COVID-19) is increasing worldwide, promising COVID-19 vaccine candidates are being produced, necessitating consideration of their potential demand, distribution, and adoption to optimize their desired effects. Objective: To assess knowledge and uptake of the Covid-19 vaccine and associated factors among adults in Shebe Sombo woreda Oromia Region South western Ethiopia, 2022 Methods: Community based cross-sectional study design was conducted from June-July, 2022 in Shebe Sombo woreda Oromia Region South western Ethiopia. A systematic random sampling technique was used in sampling the study participants. Data was checked, cleared and entered to EPI data 3.1 and exported to SPSS version 26. Then frequencies, proportions and analysis were done in bi-variable. P-value<=0.25 was used to identify candidate variables then executed multivariate logistic regression that p-value <=0.05, AOR and 95%CI were considered to identify significant variables to dependent variables 0.757-0.833. Result: A total of 395 households participated in the study 79.5% (314) (CI=0.757-0.833) had good knowledge on COVID-19 vaccination. According to this study 53.5% of participants have vaccinated for covid-19 vaccines at least once. Sex of the respondent (AOR =1.44(95%CI: 1.05, 3.92)), educational status (AOR =3.22(95%CI: 1.24, 9.86)) and residence of the respondent (AOR =1.15(95%CI: 1.02, 3.72)) were significantly associated with good knowledge of covid 19 vaccine. Knowledge of covid-19 vaccine (AOR = 1.48(95%CI: 1.08, 2.24)), residence of the respondent (AOR = 1.08(95%CI: 1.05, 1.64)), receiving any vaccination during childhood (AOR = 2.02(95%CI: 1.05, 4.92)) and having family members have been diagnosed with COVID (AOR = 1.13(95%CI: 1.03, 3.48)) were significantly associated with uptake of covid-19 vaccine. Conclusion: - In this study knowledge and uptake of covid-19 was low. Sex, educational status and residence are significant variable for good knowledge of covid-19 vaccine whereas knowledge of the respondent, received any vaccination during childhood, family members have been diagnosed with COVID-19 and residence are significant variables for uptake of Covid-19 vaccine en_US
